TB-500 is a synthetic version of the full-length 43-amino-acid peptide Thymosin Beta-4 (Tβ4), a ubiquitous intracellular protein with a central role in cytoskeletal organisation. First isolated from calf thymus extract in the 1980s, Tβ4 is present in virtually all mammalian tissues and is released by platelets and macrophages in tissue-injury models.
Its principal mechanism of action as studied in vitro involves the sequestration of G-actin (globular actin) monomers via a highly conserved LKKTET actin-binding domain, thereby modulating the ratio of monomeric to filamentous actin available for cytoskeletal remodelling. This property underpins its use as a tool in cell-migration and wound-closure assay models.
Thymosin Beta-4 has also been studied in the context of angiogenesis — specifically, endothelial cell tubulogenesis and VEGF-dependent vessel sprouting in Matrigel-based in-vitro assays. Its role in Akt kinase pathway activation has been a subject of particular interest in cardiac cell-culture research.
