SS-31 (elamipretide) is a mitochondria-targeting synthetic tetrapeptide: D-Arg-dimethyltyrosine-Lys-Phe-NH₂. The alternating cationic and aromatic amino acid architecture — characteristic of the Szeto-Schiller (SS) peptide family — confers high membrane permeability and selective accumulation within the inner mitochondrial membrane (IMM).
The compound has been extensively characterised in vitro for its binding to cardiolipin, the signature phospholipid of the IMM. This interaction stabilises cytochrome c association with the membrane, preserving electron-transport chain (ETC) architecture and reducing electron leak — a mechanism central to mitochondrial bioenergetics research.
In isolated mitochondria preparations, SS-31 has been studied for its effects on membrane potential (ΔΨm), ATP synthesis efficiency, and reactive oxygen species (ROS) generation under substrate-limited conditions, making it a widely used tool compound in mitochondrial-stress assay systems.
