Sermorelin is a synthetic 29-amino-acid peptide corresponding to the N-terminal biologically active portion of endogenous human growth-hormone-releasing hormone (GHRH) — specifically the GHRH(1-29)-NH₂ fragment. It represents the minimal sequence required for full binding and agonist activity at the pituitary GHRH receptor (GHRHR) and is used as the native-sequence reference compound in GH-axis research.
In pituitary somatotroph cell-culture models, sermorelin binds GHRHR with high affinity, activating adenylyl cyclase through Gαs coupling to elevate intracellular cAMP, which drives GH gene transcription and secretory vesicle exocytosis. It is used as the benchmark against which DPP-IV-resistant analogues (CJC-1295, tesamorelin) are compared in receptor-binding and signalling-kinetics studies.
As native GHRH(1-29) is rapidly cleaved by plasma DPP-IV at the Ala²-Asp³ bond, sermorelin has a short biological half-life in plasma-stability assays — a property studied as a baseline for evaluating modified analogues. The compound has a well-established pharmacological literature spanning several decades of GH-axis research.
