Semax is a synthetic heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) based on the ACTH(4-10) sequence — the fragment of adrenocorticotropic hormone associated with its neurotrophic rather than adrenal effects — with a C-terminal Pro-Gly-Pro extension that increases resistance to enzymatic degradation. It was developed at the Institute of Molecular Genetics of the Russian Academy of Sciences.
In neuronal cell-culture and rodent brain-tissue models, Semax has been studied for apparent upregulation of brain-derived neurotrophic factor (BDNF) and its cognate receptor TrkB, as well as effects on VEGF and NGF expression. Research has also examined its influence on dopaminergic and serotonergic signalling in synaptoneurosome preparations.
The compound has approved pharmaceutical status in Russia and is of ongoing interest in the international neuropeptide literature as a tool for studying ACTH-derived fragment pharmacology and neurotrophic factor regulation in vitro.
