Nicotinamide adenine dinucleotide (NAD⁺) is a ubiquitous dinucleotide coenzyme central to cellular oxidation–reduction (redox) reactions. It functions as a hydride-transfer carrier in glycolysis, the citric acid cycle, and oxidative phosphorylation, accepting electrons from metabolic substrates to form NADH, which donates electrons to the mitochondrial electron-transport chain.
Beyond its canonical role in energy metabolism, NAD⁺ serves as an obligatory co-substrate for sirtuin deacylases (SIRT1-7), poly(ADP-ribose) polymerases (PARPs), and cyclic ADP-ribose synthases (CD38/CD157). The relationship between NAD⁺ bioavailability and these enzyme activities has become a major research area in ageing biology and stress-response studies in vitro.
The progressive decline in NAD⁺ levels observed in ageing cell-culture models has driven substantial interest in NAD⁺ precursor and replenishment research. Experiments in yeast, C. elegans, Drosophila, and mammalian cell lines have used exogenous NAD⁺ to investigate its role in DNA repair, mitochondrial quality control, and circadian-clock regulation.
