MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA type-c) is a 16-amino-acid peptide encoded within the mitochondrial genome — specifically within the 12S rRNA gene — making it one of a class of mitochondria-derived peptides (MDPs) that function as retrograde signalling molecules between the mitochondrion and the nucleus.
First characterised by Lee et al. in 2015 (Cell Metabolism), MOTS-c is studied in vitro for its role in metabolic regulation. Research in cell-culture models has examined its apparent activation of the AMPK pathway and its effects on glucose uptake and fatty-acid oxidation in skeletal-muscle cell lines.
Of particular interest is the translocation of MOTS-c from mitochondria to the nucleus in response to metabolic stress in vitro, where it appears to interact with the antioxidant response element (ARE) promoter region of stress-response genes — a mechanism studied in the context of mitohormesis and mitochondrial–nuclear crosstalk.
