LL-37 is the only member of the cathelicidin family expressed in humans — a 37-amino-acid cationic antimicrobial peptide cleaved from the C-terminal domain of the precursor protein hCAP18. It is produced primarily by neutrophils, keratinocytes, and epithelial cells, and its name reflects the two N-terminal leucine residues and the 37-residue chain length.
LL-37 has been extensively characterised in vitro for its capacity to disrupt bacterial membranes through carpet or toroidal-pore mechanisms, with demonstrated activity against Gram-positive and Gram-negative organisms in minimum inhibitory concentration (MIC) assays. These properties make it one of the most studied human host-defence peptides in the antimicrobial literature.
Beyond direct antimicrobial activity, LL-37 is studied in vitro for innate immune signalling — specifically its ability to bind and neutralise lipopolysaccharide (LPS), modulate Toll-like receptor (TLR) activity, and act as a chemoattractant for neutrophils and monocytes in cell-migration models.
