Ipamorelin is a synthetic pentapeptide growth-hormone secretagogue (GHS) first described by Raun et al. (1998). It incorporates two non-standard residues — α-aminoisobutyric acid (Aib) at position 1 and D-2-naphthylalanine (D-2Nal) at position 3 — which together confer high metabolic stability and selectivity for the growth-hormone secretagogue receptor (GHS-R1a, the ghrelin receptor).
In pituitary cell-culture models, ipamorelin has been investigated for its dose-dependent stimulation of GH secretion. It is distinguished from earlier GHS peptides by its apparent receptor selectivity: unlike GHRP-2 and GHRP-6, which also stimulate cortisol and prolactin release in some in-vitro pituitary systems, ipamorelin is described in the literature as a selective GH secretagogue with negligible effect on these other pituitary hormones in cell assays.
This selectivity profile has made ipamorelin a widely used in-vitro tool compound in studies comparing GHS receptor subtypes, assaying GH-axis signalling cascades (MAPK, PI3K/Akt), and investigating ghrelin receptor pharmacology.
