IGF-1 LR3 (Long R3 Insulin-like Growth Factor-1) is a recombinant analogue of human IGF-1 comprising an 83-amino-acid chain with an N-terminal 13-amino-acid extension and a Glu³→Arg substitution (the "R3" modification). The R3 change substantially reduces binding affinity for IGF-binding proteins (IGFBPs) in serum, compared to native IGF-1.
The reduced IGFBP affinity means that in cell-culture and bioreactor systems, IGF-1 LR3 remains freely available to bind insulin and IGF-1 receptors on the cell surface for substantially longer than the native molecule — making it a preferred tool compound for studies of IGF-1 receptor (IGF-1R) activation, downstream PI3K/Akt/mTOR and MAPK pathway signalling, and cell-cycle progression.
IGF-1 LR3 is used extensively in bioprocess research (recombinant protein and monoclonal antibody production in CHO and HEK293 cell lines) as a cell-culture supplement, and in academic research studying IGF-1-mediated proliferation, apoptosis resistance, and proliferative and metabolic signalling in myocyte, hepatocyte, and cancer cell-line models.
