GHRP-6 (Growth Hormone Releasing Peptide-6) is a first-generation synthetic hexapeptide secretagogue structurally related to enkephalin. It was one of the compounds whose study led to the identification of the ghrelin receptor (GHS-R1a), and remains a standard reference compound in GH-axis pharmacological research.
In pituitary and hypothalamic cell models, GHRP-6 activates GHS-R1a to stimulate GH release through a phospholipase C / calcium-dependent intracellular signalling cascade. Compared to later-generation compounds such as ipamorelin, GHRP-6 has a broader receptor-activity profile — studies have reported stimulation of cortisol, prolactin, and ghrelin release alongside GH in some cell-based assay systems, making it a useful comparator compound in receptor-selectivity experiments.
GHRP-6 is also studied in hypothalamic NPY-neuron culture models for its activation of ghrelin-receptor subsets in the arcuate-nucleus region — a mechanism of interest in energy-homeostasis in-vitro research.
