GHRP-2 (Growth Hormone Releasing Peptide-2) is a second-generation synthetic hexapeptide growth-hormone secretagogue. It was developed as part of the enkephalin-derived peptide series that led to the identification of the ghrelin receptor (GHS-R1a), and incorporates D-Ala at position 1 and D-2-naphthylalanine (D-2Nal) at position 2 to improve metabolic stability relative to first-generation GHRP-1.
In pituitary cell-culture and hypothalamic tissue models, GHRP-2 activates GHS-R1a with high affinity, stimulating GH secretion via phospholipase C-mediated IP3/DAG signalling and an increase in intracellular calcium. Uniquely, GHRP-2 is also reported to exert modest effects on ACTH and cortisol release in some in-vitro pituitary models — a distinction of pharmacological interest versus more selective secretagogues.
It is widely used as a benchmark compound in GHS-R1a binding assays and receptor-selectivity studies, frequently paired with CJC-1295 (no DAC) or sermorelin in combination in-vitro experiments examining GH-axis synergy.
