CJC-1295 with DAC is a modified version of the 29-amino-acid GHRH analogue incorporating a Drug Affinity Complex (DAC) — a maleimidoproprionic acid (MPA) linker attached to the C-terminal lysine residue. The DAC technology enables covalent attachment to circulating serum albumin via reaction with endogenous thiols, dramatically extending the peptide's half-life in plasma-stability studies compared to the no-DAC form.
In GHRH-receptor binding assays, CJC-1295 with DAC retains full agonist activity and stimulates cAMP production in somatotroph cell lines with comparable potency to native GHRH. The albumin-binding mechanism is studied as a pharmacokinetic modelling tool and as a paradigm for the half-life extension approach used in therapeutic peptide design.
Researchers use CJC-1295 with DAC as a comparator compound in studies evaluating the effect of sustained GHRH-receptor exposure on GH pulsatility in pituitary cell cultures and hypothalamic–pituitary co-culture systems.
