CJC-1295 (no DAC), also known as Modified GRF(1-29) or Mod GRF 1-29, is a 29-amino-acid synthetic analogue of growth-hormone-releasing hormone (GHRH). It incorporates four amino-acid substitutions relative to native GHRH(1-29) to improve proteolytic stability against plasma dipeptidylpeptidase-IV (DPP-IV) and endopeptidase cleavage.
In pituitary somatotroph cell-culture models, the compound binds and activates the GHRH receptor (GHRHR) with high affinity, stimulating adenylyl cyclase through a Gαs-dependent pathway to elevate intracellular cAMP — the primary second-messenger for GH synthesis and release. Its improved metabolic stability extends the effective receptor-activation window compared to native GHRH(1-29) in in-vitro kinetic studies.
The compound is commonly used alongside GHS-R1a agonists (ipamorelin, GHRP-2, GHRP-6) in combination assays studying the additive or synergistic effects of combined GHRH-receptor and ghrelin-receptor stimulation on GH secretion — a research model that mimics the dual-hypothalamic control of GH release.
