Cagrilintide is a long-acting synthetic amylin analogue designed for extended receptor engagement at the amylin receptor (AMY) and calcitonin gene-related peptide receptor (CGRP-R). It incorporates a fatty-acid side chain modification that confers prolonged albumin binding and an extended pharmacokinetic profile compared to native amylin.
In vitro, cagrilintide has been studied for receptor binding at AMY1-3 (amylin receptor subtypes comprising calcitonin receptor with RAMP subunits) and CGRP-R, both Gαs-coupled GPCRs. Cell-based assays have examined its Gαs/cAMP signalling and β-arrestin recruitment profile in recombinant receptor systems.
