Ara-290 (cibinetide) is an 11-amino-acid synthetic peptide derived from the helix-B surface peptide (HBSP) region of erythropoietin (EPO). Unlike EPO itself, Ara-290 is engineered to bind the tissue-protective, non-haematopoietic receptor subtype — the innate repair receptor (IRR), a heteromeric complex involving the β-common receptor (βcR) — rather than the classical EPO receptor homodimer responsible for erythropoiesis.
In vitro, the compound has been studied for its interactions with the IRR and for effects on downstream signalling pathways including JAK2/STAT3 and PI3K/Akt, which are implicated in cytoprotective and anti-apoptotic responses in neuronal and pancreatic β-cell culture models.
Ara-290 is used as a tool compound in research exploring the dissociation of EPO's haematopoietic and non-haematopoietic receptor pharmacology — a topic of growing interest in neuroprotection and metabolic-disease in-vitro research.
